ABSTRACT

Diabetic retinopathy (DR) is a common complication of diabetes, which causes a great threat to the health of human vision. Recent studies have shown that saponins, polysaccharides and flavonoids in Astragali Radix regulate phosphoinositide 3-kinase/protein kinase B/mammalian target of rapamycin pathway, silent information regulator 1/nuclear factor erythroid 2-related factor 2 pathway, B-cell lymphoma-2 and other signaling pathways by acting on nuclear factor-κB, hypoxia-inducible factor-1α, glial fibrillary acidic protein, glutamine synthetase, aldose reductase, multiple miRNA and other related targets, so as to exert anti- inflammatory, anti-microangiopathy, anti-neurodegenerative disease and anti-apoptosis, and then achieve the purpose of treating DR.

ABSTRACT

After the compatibility of Astragali Radix and Atractylodis Macrocephalae Rhizoma, most of the effective components of Astragali Radix increase, and the bioavailability is improved. Compared with the application of the two drugs alone, it can enhance the effects of immune regulation, anti-tumor, diuresis, lung protection, regulation of flora, and intestinal protection. However, the optimal compatibility ratio of Astragali Radix- Atractylodis Macrocephalae Rhizoma pair to exert various pharmacological effects still needs to be clarified. The drug pair and related preparations are mostly used in the treatment of nephropathy, but its mechanism of action needs to be further elucidated.

ABSTRACT

Objective:To investigate the medicinal plant resources in Saertu District, Heilongjiang Province so as to provide scientific foundation for the protection rational development and utilization of wild medicinal plant resources in this region.Methods:According to the technical specification requirements of the fourth national census of Traditional Chinese Medicine (TCM) resources, the wild medicinal plant resources in Saertu area were analyzed.Results:A total of 36 samples were completed, in this survey among which contains a total of 166 species of wild medicinal plant resources, belonging to 118 genera of 45 families, including 37 species of key medicinal materials.Conclusion:The TCM resource in Saertu is relatively scarce, and the species and resource distribution of TCM in this region have been basically found out, which can provide reliable information for the establishment of TCM resource protection zones in Saertu and expanded to the entire province.

ABSTRACT

OBJECTIVE To screen t he active component s of Euchresta japonica against nasopharyngeal carcinoma. METHODS Main chemical components of E. japonica were selected ,and their target proteins were predicted in Swiss Target Prediction database. The target proteins of nasopharyngeal cancer were obtained with GeneCards database. Protein-protein interaction(PPI)network was established after the target of chemical components of E. japonica was intersected with the target of nasopharyngeal carcinoma ;PPI network was analyzed by using Cytoscape 3.6.1 software,and the potential active components and key targets of E. japonica against nasopharyngeal carcinoma were screened. The molecular docking technology was used to evaluate binding ability of active component-key target ;active components of E. japonica against nasopharyngeal carcinoma were screened. The anti-nasopharyngeal cancer effect of potential active components of E. japonica was verified by cell proliferation experiment. RESULTS Seven potential active components (tonkinensisol,quercetin,sophoranone,matrine,genistein,coumarin,maackiain) and 10 core targets (SRC,PIK3CA,MAPK1,MAPK3,AKT1,MAPK8,MAP2K1,PTK2,EGFR,JAK3)of E. japonica against nasopharyngeal carcinoma were screened. The molecular docking results showed that above potential active components all possessed certain anti-nasopharyngeal cancer effect. Cell proliferation activity test showed that tonkinensisol ,sophoranone and maackiain had a very significant inhibitory activity on nasopharyngeal carcinoma cells CNE- 1. CONCLUSIONS Tonkinensisol, sophoranone and maackiain might be the main active components of E. japonica against nasopharyngeal carcinoma.

ABSTRACT

Objective:To establish a qualitative and quantitative analysis method of ginsenoside Rb1, cinnamic acid, paeoniflorin and berberine in Wenxin formula based on UPLC-MS/MS MRM model so as to provide a rapid and accurate evaluation method for the research and development of new drugs.Methods:Adopting Waters ACQUITY UPLCTM HSS T3 (2.1 mm×100 mm, 1.8 μm) and gradient elution was performed by mobile methanol-0.03% formic acid water. chromatographic column was used with electrospray ionization source in the scanning mode of multiple reactive ion monitoring (MRM) for ion separation and screening. The retention time and the relative abundance ratio (qualitative ion pairs/quantitative ion pairs) was used for qualitative analysis, while quantitative ion pairs was used for quantitative analysis.Results:The four components tested in Wenxin Formula have been qualitatively detected and showed a good linear relationship, the method showed accuracy, precision, repeatability, and stability, the recovery rate was 96.24%-104.19% and RSD was 1.29%-3.30%. Conclusion:The established method is simple, accurate, rapid and sensitive, which is reliable for the qualitative and quantitative study of the four main components in Wenxin Formula.

ABSTRACT

Objective:To analyse the material basis and potential mechanism of Acanthopanax senticosus injection (ASI) in the treatment of ischemic stroke by combining UPLC-Q/TOF-MS and network pharmacology.Methods:The chemical composition of ASI was identified by UPLC-Q/TOF-MS. The Swiss Target Prediction, GeneCards and OMIM databases were used to predict the potential targets for the action of ASI in the treatment of ischemic stroke. The String database and Cytoscape software were used to construct protein interaction network maps, and the Omicshare platform was used to perform gene ontology (GO) and KEGG enrichment analysis. The DockThor platform was used for molecular docking.Results:The analysis of 53 components in ASI was firmly established and used as a basis to obtain 189 related targets of ASI for the treatment of ischemic stroke. Reverse screening showed that 25 components in ASI may be important active components in the treatment of ischemic stroke. Functional enrichment studies found that ASI may mainly regulate PI3K-Akt signaling pathway to treat ischemic stroke.Conclusion:This study preliminarily predicted the mechanism of ASI in the treatment of ischemic stroke may be related to inhibition of inflammation, antioxidant stress, promotion of angiogenesis and protection of nerve cells.

ABSTRACT

Sijunzi decoction (SJZD) is a Chinese classical formula to treat spleen qi deficiency syndrome (SQDS) and has been widely used for thousands of years. However, the quality control (QC) standards of SJZD are insufficient. Chinmedomics has been designed to discover and verify bioactive compounds of a variety of formula rapidly. In this study, we used Chinmedomics to evaluate the SJZD's efficacy against SQDS to discover the potential quality-markers (q-markers) for QC. A total of 56 compounds in SJZD were characterized in vitro, and 23 compounds were discovered in vivo. A total of 58 biomarkers were related to SQDS, and SJZD can adjust a large proportion of marker metabolites to normal level and then regulate the metabolic profile to the health status. A total of 10 constituents were absorbed as effective ingredients that were associated with overall efficacy. We preliminarily determined malonyl-ginsenoside Rb2 and ginsenoside Ro as the q-markers of ginseng; dehydrotumulosic acid and dihydroxy lanostene-triene-21-acid as the q-markers of poria; glycyrrhizic acid, isoglabrolide, and glycyrrhetnic acid as the q-markers of licorice; and 2-atractylenolide as the q-marker of macrocephala. According to the discovery of the SJZD q-markers, we can establish the quality standard that is related to efficacy.

A canthopanax Senticosus , a vulnerable species, was one of the major government-protected plants. This paper deals with the major reason for wildlife resources from biological standpoints, which are sexual propagation with less capability , vegetative propagation being damaged due to leaves , roots and rhizomes being gathered un-reasonable. Raising wildlife resources, semi-wild extensive management, and reasonable gatheration will be an important means for protection of wildlife resources in light of local conditions.

Traditional Chinese medicine (TCM) has been used to prevent disease for thousands of years. However, the actual value of TCM has not been recognized adequately due to the lack of scientific approaches to its study . Fortunately, metabolomics becomes practically available and consists with the holistic thinking of TCM. The metabolomics can serve as a new strategy for evaluating the efficacy of TCM . This review summarized applications of metabolomics in special TCM field with an emphasis on biomarker discovery.

ABSTRACT

Serum pharmacochemistry of traditional Chinese medicine (TCM) is a direct and effective method for determining efficacious substance foundation of TCM. However, the complexity of chemical constituents of TCM and the interaction among ingredients in the in vitro process make the analysis on in vitro ingredients of TCM arduous. Liquid chromatography coupled with tandem mass spectrometry (LC-MS/MS) has become the cornerstone in detection and characterization of in vivo ingredients of TCM because of its sensitivity and ability to analyze complex mixtures. However, due to significant interference from endogenous species, detection and identification of the constituents of TCM in the biological matrices are often difficult. There is a crying need for introducing specialized ingredient identification techniques to avoid artificial omission of in vivo ingredients of TCM. On the basis of the analysis on transitional ingredients in rat blood, this essay introduces the application of such pattern recognition methods as mass defect filter, Metabolynx software and principal component analysis, partial least squared discriminant analysis and orthogonal partial least squared discriminant analysis in identifying in vivo ingredients of TCM.

ABSTRACT

The origin place change is an almost universal rule for many Chinese medicines according to the formation and change history of ancient and recent Dao-di herbs. Among them, the quality and output are the essence and driving force of the formation and change of Dao-di herbs origin place. More and more species and quantities of Chinese medicine were cultivated along with development of Chinese medicine and modern manufacturing technology, and its production regions far exceed natural location zones, the rationality of origin place change should be based on drug quality evaluation. Under the guidance of the formation mode of Dao-di herbs, characteristics of Dao-di herbs were summarized, its characteristics and applied limits were suggested, some traditional Chinese medicinal materials only with higher output, cultivated materials in distribution area for wild, or materials with poor genetic resources failing to answer with Dao-di herbs. Chinese medicines authenticity should strengthen the study of germplasm and drug quality assessment, the suitability of producing area should be assessed according to output and quality under the guidance of suitable locations regionalization.

ABSTRACT

By using UPLC coupled with MS, the analysis of the constituents absorbed into blood after oral administration of Yinchenhao decotion with different combination had been carried out, the body fate of constituents detected in the blood was elucidated also; At the same time, the overall body respones after multi-input of the medical formule were evaluated by metabolomic analysis. These data explained the scientific values in both drug metabolisim and metabonomics, the results had given some new evidences for the potentialities of pharmacometabonomics to elucidate the compatibility principle of Chinese medical formule. As a conclusion, concept of "Pharmacometabolomics of Chinmediformulae" has been defined.

ABSTRACT

Objective To develop a method for simultaneous determination of 4 compounds in Zhizhu Pills by HPLC.Method Reversed-phase HPLC was used to determine the content of the compounds such as hesperidin,naringin,neohesperidin and atractylenolideⅠ.The column was symmetry C18(250 mm?4.6 mm,5 ?m) with temperature of 30 ℃.Methanol-0.095 %phosphoric acid solution served as the mobile phase by gradient elution.The wavelength of detection was set at 283 nm and 224 nm.The injection volume was 10 ?L and the flow rate was 1.0 mL/min.Results Hesperidin,naringin,neohesperidin and atractylenolideⅠin Zhizhu Pills can be isolated completely.Three batches of samples were determined and the recoveries of naringin,hesperidin,neohesperidin and atractylenolideⅠwere 98.81 %(RSD=1.11 %,n=6),100.63 %(RSD=1.90 %,n=6),99.32 %(RSD=1.44 %,n=6) and 99.65 %(RSD=1.53 %,n=6) respectively.Conclusion This method is accurate,reliable and with good reproducibility.It can be used for the quality control of Zhizhu Pills.

ABSTRACT

Objective To determine the content of two original constituents of Liuwei Dihuang Wan migrating to blood. Methods HPLC- fingerprint of the preparation and drug- containing serum fingerprint were established by RP- HPLC. Based on the fingerprints, the original constituents migrating to blood were identified and determined by comparing with the standard. Results Loganin and paeonol were identified as the original constituents migrating to blood; the loganin content was 0.93? 0.01 mg/g and the paeonol was 0.37? 0.01 mg/g. Conclusion Using the constituent content migrating to blood as the parameter, this method will provide scientific evidence for improving the quality control of Liuwei Dihuang Wan. 

Their activity rate of steady ClO2 was measured under different pH condition. Results With the decrease of pH,the activity rate of steady ClO2 increased. Aim To establish a set of consummate evaluation methods of animal models which are with syndromes of traditional Chinese medicine.Method By means of 2,4-dinitrophenol-induced fever syndrome model for the entry point,making use of meta bonomics as the platform,through analyzing metabolic fingerprint data of rat urine in the control and model group and metabolome of rat urine in model group at different time intervals,to approach the evaluation methods of animal models.Results Through research by metabonomics,the results showed that 2,4-dinitrophenol-induced animal models consisted with clinical fever syndrome of traditional Chinese medicine.Conclusion Metabonomics can be used for evaluation studies of animal models which are with syndromes of traditional Chinese medicine.
